{"id":42041,"date":"2022-02-07T07:25:05","date_gmt":"2022-02-07T07:25:05","guid":{"rendered":"https:\/\/i-base.info\/htb\/?p=42041"},"modified":"2022-02-09T18:32:30","modified_gmt":"2022-02-09T18:32:30","slug":"drug-resistance-profile-of-integrase-inhibitors-when-treating-hiv-2","status":"publish","type":"post","link":"https:\/\/i-base.info\/htb\/42041","title":{"rendered":"Drug resistance profile of integrase inhibitors when treating HIV-2"},"content":{"rendered":"<div>\n<p class=\"HTBBODYtext\"><strong>Simon Collins, HIV i-Base<\/strong><\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\"><strong><span lang=\"EN-US\">Treatment guidelines for HIV-1 have for many years recommended integrase inhibitor-based ART as first-line ART.<\/span><\/strong><\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\"><span lang=\"EN-US\">A study just published in JID includes an analysis of drug resistance mutations that are specific to HIV-2, with significant loss in phenotypic sensitivity.<\/span><\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\"><span lang=\"EN-US\">These will be an essential reference for optimal management of HIV-2 where maintaining sensitivity to integrase inhibitors will be essential for long-term care.<\/span><\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\"><span lang=\"EN-US\">The abstract results note: &#8220;<\/span>We observed extensive cross-resistance between raltegravir and dolutegravir in HIV-2<sub>ROD9<\/sub>. HIV-2\u2013specific integrase mutations Q91R, E92A, A153G, and H157Q\/S, which have not been previously characterised, significantly increased the EC<sub>50<\/sub>\u00a0for raltegravir when introduced into one or more mutational backgrounds; mutations E92A\/Q, T97A, and G140A\/S conferred similar enhancements of dolutegravir resistance. HIV-2<sub>ROD9<\/sub>\u00a0variants encoding G118R alone, or insertions of residues SREGK or SREGR at position 231, were resistant to both INIs.&#8221;<\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\">Reference<\/p>\n<\/div>\n<div>\n<p class=\"HTBBODYtext\"><span lang=\"EN-US\">Smith RA et al for the University of Washington-Senegal HIV-2 Study Group.
