Etravirine interactions

A comprehensive review by Tibotec colleagues of the pharmacokinetic interaction studies between the NNRTI etravirine and non-antiretroviral drugs.
The data are summarised in tables (least square means ratio of AUC, Cmax and Cmin of either etravirine or coadministered drug) with references. In addition, recommendations are given in relation to dose adjustments. The potential for interaction with some key drugs (eg ribavirin, opioid analgesics, dexamethasone, calcineurin inhibitors, antiepileptics) not yet formally studied is presented based on metabolic profiles.
Although the authors conclude that the potential for interactions between etravirine and non-antiretroviral agents is limited there are some clinically relevant interactions to note:
Not recommended: Antiepileptics (carbamazepine, oxcarbazepine, phenobarbitone, phenytoin), rifampicin, rifapentine, St Johns wort.
Consider alternatives: Alternatives to clopidogrel or diazepam should be considered.
